| New, on-line glucose measurements of 0.01 - 1 g/L |
|
Braunschweig, August 1st, 2005 - Glucose is by far the most important substrate for microorganisms in biotechnology processes. It is used as carbon source in almost 95% of all cultivations. Control of the glucose concentration can be required by the process or is simply recommended for economical reasons. A typical example is the high cell density cultivation of E. coli for the production of heterologous proteins. The glucose concentration must remain below 1 g/l during the production phase of this fed-batch process to prevent the formation of unwanted metabolites. On the other side, total depletion of the glucose will immediately lead to a decrease in cell viability. Thus, the control of the substrate feed requires a timely and reliable measurement of the present concentration. Analysis methods like HPLC or enzyme kits are capable of measuring these glucose concentrations, but are relatively expensive or problematic for on-line analysis. ProcessTRACE allows a rapid and precise determination of the glucose concentration in the fermenter and provides the tools for a quick set-up of a feed control strategy.
For the optimized dosage of glucose, the new application has been developed in the limited range of 0.01 - 1 g/L.
